• MnFe2O4/MoS2 nanocomposites were prepared by a sonochemical method. • MoS2 nanosheets were exfoliated and decorated homogeneously with MnFe2O4 nanoparticles. • Mesoporous structure with specific surface area of 97.16 m2/g. • Superparamagnetic behavior with saturation magnetization of 37.4 Am2/kg.<br>This study established a facile one-step strategy to anchor MnFe2O4 nanoparticles on the surface of MoS2 nanosheets in a controlled manner. The as-prepared MnFe2O4/MoS2 nanocomposites were investigated by TEM, XRD, XPS, Raman, BET and VSM in detail. The MnFe2O4 nanoparticles with an average particle size of 26 nm were densely and uniformly decorated on MoS2 nanosheets, and consequently, both the aggregation of MnFe2O4 nanoparticles and restacking of MoS2 nanosheets were effectively prevented. More importantly, the MnFe2O4/MoS2 nanocomposites exhibited high specific surface area, typical superparamagnetic behavior and excellent solution dispersion, showing a great potential for biomedical applications in the fields of magnetic resonance imaging, targeted drug delivery and tumor hyperthermia.

Rheum tanguticum Maxim. ex Balf. (Rt), a traditional Tibetan medicine, is known to exert various bioactivities, including anti-inflammatory and antioxidative activities. The present study was conducted to investigate anti-inflammatory and antioxidative effects of Rt on activated microglia. Rt (10 μg/ml) significantly inhibited the mean protein level of interleukin-1β (IL-1β) in the organotypic hippocampal slice cultures following treatment with chromogranin A (CGA, 10 nM) and pancreastatin (10 nM), endogenous microglial activators present in senile plaques. Rt also significantly inhibited the expression and production of inflammatory and oxidative molecules, including IL-1β, tumor necrosis factor-α, and nitric oxide, by cultured microglia after treatment with CGA. These effects of Rt are considered to be mediated by the secretion of interleukin-10 (IL-10) from microglia, because neutralizing antibodies against IL-10 significantly canceled these effects. To explore the causative components of Rt responsible for inducing the secretion of IL-10, the effects of seven components of Rt on the IL-10 expression in microglia were examined. Among them, aloe-emodin (10 μM) and (+)-catechin (30 μM) were able to induce the secretion of IL-10 from cultured microglia. Therefore, aloe-emodin and (+)-catechin are deemed responsible for the antineuroinflammatory and antioxidative effects of Rt through the secretion of IL-10 from microglia. Accordingly, Rt is considered potentially useful for the treatment of AD.

A simple and sensitive high-performance liquid chromatographic (HPLC) method with fluorescence detection and mass spectrometric identification has been developed for analysis of 30 long-chain and short-chain free fatty acids (FFAs). The fatty acids were derivatized to their esters with 1-[2-(<i>p</i>-toluenesulfonate)ethyl]-2-phenylimidazole-[4,5-<i>f</i>]-9,10-phenanthrene (TSPP) in <i>N</i>,<i>N</i>-dimethylformamide (DMF) at 90 °C with anhydrous K<sub>2</sub>CO<sub>3</sub> as catalyst. A mixture of C<sub>1</sub>-C<sub>30</sub> fatty acids was completely separated within 60 min by gradient elution on a reversed-phase C<sub>8</sub> column. Qualitative identification of the acids was performed by atmospheric-pressure chemical ionization mass spectrometry (APCI-MS) in positive-ion mode. The fluorescence excitation and emission wavelengths were 260 and 380 nm, respectively. Quantitative determination of the 30 acids in two Tibetan medicines <i>Gentiana straminea</i> and <i>G. dahurica</i> was performed. The results indicated that the medicines contained many FFAs. Linear correlation coefficients for the FFA derivatives were >0.9991. Relative standard deviations (RSDs, <i>n</i> = 6) for the fatty acid derivatives were <3%. Detection limits (at a signal-to-noise ratio of 3:1) were 3.1-38 fmol. When the fatty acid derivatives were determined in the two real samples results were satisfactory and the sensitivity and reproducibility of the method were good.

Abstract This presented study describes a method based on high performance liquid chromatography combined with fluorescence detection (HPLC-FLD) using N-(2-iodoacetyl)-1-pyrenemethylamine (NIPA) as a novel fluorescence labeling reagent for the determination of thyreostats in bovine milk. Five thyreostats, belonging to the group of imidazole and thiouracil, were investigated in this work: tapazole (TAP), thiouracil (TU), methylthiouracil (MTU), propylthiouracil (PTU) and phenylthiouracial (PhTU). Thyreostats were specifically purified by a silver ion solid phase extraction (Ag-SPE) cartridge and then labeled using NIPA. The labeled derivatives showed excellent fluorescence property with maximum excitation and emission wavelengths of 330 nm and 375 nm, respectively. The labeled derivatives were separated on a reversed-phase Eclipse SB-C18 column within 12 min. Excellent linearity (R2 > 0.995) of all thyreostats was achieved with the limits of detection (LODs) and the limits of quantitation (LOQs) in the low micrograms per liter range of 0.21–0.30 μg/L and 0.70–1.00 μg/L, respectively. Satisfactory recoveries in the range of 93.5–98.0% were obtained for all thyreostats. The developed method has been successfully applied to analyze thyreostats in bovine milk with good applicability. Thirty bovine milk samples have been investigated, and varying levels of thiouracil were detected in thirteen of these samples. The highest level in the raw milk reached a value of 4.5 μg/L. To our best knowledge, this study is the first to report the presence of naturally occurring thiouracil in milk by HPLC-FLD analysis. Chemical isotope labelling in combination with high-performance liquid chromatography-tandem mass spectrometry (CIL-HPLC-MS/MS) is a powerful method for quantitative profiling of targeted molecules. In the current work, we successfully developed a novel CIL-HPLC-MS/MS method for quantitative profiling of residual organophosphorus thioester pesticides (OPTPs) in agricultural products through the determination of the cleavage products of thiol (CP-thiol) compounds. In this method, we synthesized a novel pair of CIL reagents, i.e., N-(4-(carbazole-9-yl)-phenyl)-N-maleimide (NCPM-d0) and its deuterated analogue NCPM-d2, both of which contain a maleimide moiety as the reactive group and an isotope tag to sensitively label CP-thiol compounds. NCPM-d0 was used to label CP-thiol compounds cleaved from OPTPs in the investigated agricultural product samples, and NCPM-d2 was used to label CP-thiol compounds cleaved from OPTPs in the standard substance-spiked organic agricultural product samples. The heavily labelled derivatives were used as the internal standards (ISs) to compensate for the matrix effects during MS analysis. The NCPM-d0- and NCPM-d2-labelled derivatives generated two characteristic product ions (PIs) at m/z 372.5 and 374.5 under collision induced dissociation, respectively, which are used to establish the multiple reaction monitoring (MRM) mode-based detection. The precursor ions of NCPM-d0 and NCPM-d2 labelled derivatives of CP-thiol compounds were deduced according to the structures of the OPTPs. The peak pairs with a fixed mass difference and similar retention times were assigned as potential CP-thiol candidates for the identification of the corresponding OPTPs. Using the proposed method, we successfully determined seven residual OPTPs in agricultural product samples. Taken together, the presented method was demonstrated to be a promising new technique in the quantitation of OPTPs in agricultural product samples with high reliability.

This study is aimed to establish a high-performance liquid chromatography (HPLC) method for simultaneous determination of skimmin, scopolin and umbelliferone in Saussurea hieracioides. Samples were analyzed on a Wondasil C18-WR column (4.6 mm x 250 mm, 5 microm) with methanol (A) and water containing 0.1% phosphate (B) as mobile phases for gradient elution at a flow rate of 1.0 mL x min(-1). The detection wavelength and column temperature were set at 325 nm and 35 degrees C, respectively, and the sample size was 10 microL. The results showed that skimmin, scopolin and umbelliferone were simultaneously achieved within 40 min under the above conditions. A good linearity was observed in the range of 0.18-5.6 microg (r = 1.000 0), 0.060-1.8 microg (r = 0.999 9), 0.032-0.97 microg (r = 0.999 8) for skimmin, scopolin and umbelliferone, respectively, with the average recoveries of 99.16% (RSD = 0.41%), 100.3% (RSD = 0.79%), 102.2% (RSD = 0.87%). The method is simple, accurate and reproducible and can be used for the quality control of S. hieracioides.

An improved HPLC-DAD-ESI-MS(n) method has been developed to simultaneously quantify eight major compounds in Saussurea tridactyla Sch.-Bip. ex Hook. f. which has long been used as a traditional Tibetan medicine. This method was validated to be sensitive, precise and accurate with the LODs of 0.11-5.01 microg/ml, the overall intra-day and inter-day variations less than 2.70%, and the overall recovery over 98.0%, respectively. The correlation coefficients (r(2)) of the calibration curves were higher than 0.991. This newly established method was successfully applied to reveal the difference in the chemical profiles and contents of these analyses in S. tridactyla from different localities. In addition, by comparison UV and MS spectra with those of authentic compounds and literatures, a total of fourteen peaks were identified. It can be concluded that this method was effective to ensure the safety and efficacy consistency of S. tridactyla, and can be applied to other traditional Tibetan medicinal plants from different resources in Tibet.

OBJECTIVE: To establish the HPLC fingerprint for Halenia elliptica herbs, a traditional Tibetan medicine, in order to study constituents contained in H. elliptica from different habitats and compare their differences.METHOD: HPLC analysis was made on a Welchrom-C18 (4.6 mm x 250 mm, 5 microm) with water and acetonitrile as mobile phase. The wavelength was detected as 265 nm, the flow rate was 1.0 mL x min(-1), and the column temperature was 40 degrees C. The software for chromatographic fingerprint was applied to analyze the similarity. And principal component analysis was conducted. RESULT: Twelve common chromatographic peaks were identified by fingerprint, showing a low similarity in constituent and variety. The significant difference in the proportion between xanthones and aglycones in each batch of herbs indicated no notable correlation between constituent characteristics and geographic locations of habitats. CONCLUSION: The method is so simple, exclusive, stable and highly repeatable that it can provide reference for identification and quality assessment of H. elliptica herbs.

Soil respiration (Rs) is an important source of atmospheric CO2 flux and is sensitive to changes in soil nutrient and water contents. Despite extensive studies on the effects of enhanced atmospheric nitrogen (N) deposition and changes in precipitation (P) on Rs, few studies have taken into account the effects of interactions between these factors on Rs of alpine grasslands. To address these questions, we investigated the effects of N addition (10 g N m-2 yr-1), changes in precipitation (±50% precipitation), and their interaction on soil respiration and its components, including heterotrophic respiration (Rh) and autotrophic respiration (Ra),in a Tibetan alpine steppe during three consecutive growing seasons. We found that Rs differed in its response to N addition and precipitation regimes. Specifically, decreased precipitation led to a significant reduction in Rs during the last two years, whereas N addition minimally impacted Rs. Another important finding was that soil respiration components differed in their response to N addition and precipitation regimes. Nitrogen addition significantly enhanced Ra, whereas Rh was not altered in response to N addition. By contrast, the precipitation regime led to marked changes in Rh, but exhibited marginally significant effects on Ra. Therefore, our findings highlighted that soil respiration differed in its response to N addition and precipitation regimes mainly due to the different responses of soil respiration components to these factors. Therefore, carbon dynamics should take soil respiration components into account under global change scenarios.

Western blot, immunohistochemistry and Q-PCR assay were used to determine the levels of protein and mRNA. Masson Fontana staining was used for melanin staining and FITC-Phalloidin staining was used to detect the expression of F-actin. Zebrafish and B16F10 cells were used for the mechanism research.<br>Results: Fluoxetine (2.6 mg/kg, ig) ameliorated hypopigmentation induced by CUMS and CRS in mice, significantly increased the mRNA and protein levels of 5-HT1 A and 5-HT2 A receptors in mice and B16F10 cells. The effect of fluoxetine on melanogenesis in B16F10 cells and zebrafish were inhibited by WAY100635 (a selective 5-HT1 A receptor antagonist) and ketanserin (a 5-HT2 A receptor antagonist), respectively. Activation of p38 MAPK signaling pathways was contributed to fluoxetine induced melanogenesis and inhibited by WAY100635, but not ketanserin. However, ketanserin selectively weakened the action of fluoxetine promoted migration and up-regulated Rab27a protein expression in B16F10 cells.<br>Conclusions: 5-HT1 A and 2 A receptors contribute to melanogenesis and migration property of fluoxetine. The newly revealed mechanism indicates that fluoxetine and its analogues may be a potential drug for treatment of depigmentation disorders.

ETHNOPHARMACOLOGICAL RELEVANCE: Dioscorea zingiberensis C. H. Wright (D. zingiberensis), Dioscoreaceae, is used extensively in traditional Chinese medicines. The aim of the current review paper is to give a comprehensive overview of the traditional usage and phytochemistry of the plant. Clinical studies performed and products prepared from the plant and active principles will be mentioned. In addition a review of the taxonomy of the genus Dioscorea is given.MATERIALS AND METHODS: A systematic search on literature has been performed in databases like Google Scholar, Science Direct, Scifinder, Web of Science, and in Chinese databases (China Knowledge Resource Integrated, i.e., CNKI and Wanfang) including Ph.D. and M.Sc. dissertations. These from the period 1983-2016 have been searched. RESULTS: D. zingiberensis is an endemic plant in China widely distributed in some provinces, especially Hubei and Shaanxi. The rhizomes are the medicinal part in Dun-Ye-Guan-Xin-Ning tablets and are used for isolation of diosgenin. Steroidal saponins are believed to be the active principles. More than 70 compounds have been identified. Several of these have been tested in preclinical assays and clinical trials. A wide spectrum of biological effects including cardiovascular, anti-thrombosis, hyperlipidemia, neuroprotection, anti-inflammatory, and anthelmintic effect has been verified. CONCLUSIONS: Because of the promising results from the investigations on the plant material of D. zingiberensis, further in depth analyses ought to be performed to evaluate its potential as either a traditional drug or a source of bioactive principle. The presently performed studies do not explain mechanism of action, pharmacokinetics (ADME properties), or toxicity. All of these topics need more elaborate investigations.

Although large amounts of soil organic carbon (SOC) stored in the shrublands, information about SOC storage was little on the Tibetan Plateau. This study aims to evaluate the spatial patterns and storage of SOC in the shrublands and the relationships of climatic variables and soil pH on the Tibetan Plateau.<br>We used 177 profiles of soil samples obtained from 59 shrubland sites on the northeast Tibetan Plateau from 2011 to 2013. Ordinary least squares regressions, curve estimation, and multiple linear regressions were used to evaluate controlling factors on SOC stock. Kriging interpolation was used to upscale sit-level measurements to the whole study area.<br>We found that SOC storage in the northeast Tibetan shrublands was 1.36 Pg C in the top 1 m with an average SOC stock of 12.38 kg m<sup>−2</sup>. SOC stock decreased from east to west and south to north but generally increased significantly with the mean annual temperature (MAT) and the mean annual precipitation (MAP), and tended to decrease with soil pH. Although similar relationships were also observed in alpine shrublands, the trends among SOC stock, MAP, and MAT were not observed in desert shrublands. Our results indicate that a reduction in soil pH accelerates the C sequestration potential. Furthermore, global warming contributed to C sequestration in alpine shrublands, specifically, SOC stock increased 8.44 kg m<sup>−2</sup> with an increased unit of MAT in alpine shrublands just considering temperature effects. Meanwhile, the C sequestration was different among different regions due to the uneven increases in precipitation. However, in desert shrublands, MAP and MAT did not significantly affect SOC stock.<br>The results indicate that though a reduction in soil pH could contribute to C sequestration, MAT and MAP have different effects on SOC stock in different Tibetan Plateau shrublands. Increased MAT and MAP were 0.05 °C and 1.67 mm every year on the Tibetan Plateau, which will increase C sequestration in alpine shrublands, but might have limited impacts on desert shrublands, which help us comprehend soil C cycling in the global climate change scenario.

Background Rheum tanguticum Maxim. ex Balf is one of the plants generically known as rhubarb, a culinary vegetable that has long been used as a herbal remedy both in China and Europe. Increasing demand for rhubarb has triggered the overexploitation of Rh. tanguticum. Cultivation is therefore necessary for quality control and protection of wild resources. Nitrogen fertilizer plays an important role in cultivation. This study aimed to explore how nitrogen fertilizer affects the growth and quality of rhubarb on the Qinghai-Tibetan plateau. Results Nitrogen fertilizer promoted growth but had no significant influence on the active compounds of Rh. tanguticum. Generally, the N2 (150 kg ha(-1)) and N3 (225 kg ha(-1)) levels showed the most improved growth indexes, with no significant differences between them. The growth index and the amounts of eight of the nine studied active compounds in Rh. tanguticum increased from each year to the next and differed among growth stages. The contents of the active compounds were higher at the green stage and lower at the growth stage, which was opposite to the seasonal trends in root dry matter ratio. Gallic acid levels decreased with the growth of the plant. Conclusion The N2 level (150 kg ha(-1)) was the recommended nitrogen fertilizer level in this study. It was revealed that seasonal changes rather than nitrogen fertilizer influenced active compounds in the root of Rh. tanguticum. (c) 2018 Society of Chemical Industry

A total of 253 participants were recruited for four experiments. In Study 1, the mindful-learning group reported greater levels of pro-environmental behavioral intentions compared to a randomized control. In Study 2, we utilized different learning materials focusing on self, humans, or the biosphere in three sub-experiments. Study 2a manipulated mindsets by a self-related focus and revealed participants in a mindfulness condition had lower pro-environmental behavioral intentions than those in the mindlessness group. Study 2b centered on humans and results showed that participants in a mindfulness group reported higher levels of pro-environmental behavioral intentions. Finally, Study 2c induced mindsets with a biospheric focus, showing participants in the mindful-learning condition had greater pro-environmental behavioral intentions. Combined, the studies provide empirical evidence that mindful learning could influence self-reported pro-environmental behavioral intentions both positively and negatively.

Mindfulness-based stress reduction (MBSR) has been proved to be effective in reducing stress and rarely studies were focused on Chinese pregnant women. The aim of the current paper is to investigate the effects of 8-week MBSR on prenatal stress, anxiety and depression among Chinese pregnant women. A sample of 66 pregnant women randomly allocated into either the MBSR group (n=34) or the control group (n=32). Participants in the MBSR group received a group 8-week, 90-min each time intervention. The results found a significant interaction between time and condition for prenatal stress (F=45.51, p<0.001, η 2=0.427), anxiety (F=19.30, p<0.001, η 2=0.240), while depression showed no time-by-group interaction (F=0.29, p=0.589, η 2=0.005). As for the sub-scale of state anxiety, while there was only no time effect (F=3.68, p=0.060, η 2=0.057). The findings of this study preliminary indicated effects of the MBSR intervention on self-reported prenatal stress and anxiety in comparison to a treatment-as-usual control. Effect on depression was not observed may due to the low level of depression of participants. The aim of the current paper is to investigate the effects of 8-week MBSR on prenatal stress, anxiety and depression among Chinese pregnant women. A sample of 66 pregnant women randomly allocated into either the MBSR group (n=34) or the control group (n=32). Participants in the MBSR group received a group 8-week, 90-min each time intervention. The results found a significant interaction between time and condition for prenatal stress (F=45.51, p<0.001, η 2=0.427), anxiety (F=19.30, p<0.001, η 2=0.240), while depression showed no time-by-group interaction (F=0.29, p=0.589, η 2=0.005). As for the sub-scale of state anxiety, while there was only no time effect (F=3.68, p=0.060, η 2=0.057). The findings of this study preliminary indicated effects of the MBSR intervention on self-reported prenatal stress and anxiety in comparison to a treatment-as-usual control. Effect on depression was not observed may due to the low level of depression of participants. This study provides preliminary evidence that MBSR is suitable for Chinese pregnant women and be effective in decreasing prenatal stress, anxiety.

OBJECTIVE: To explore the effects of traditional Tibetan medicine, Fructus Lonicerae microphyllae (FLM) on phagecytosis and cytokines production of murine macrophages.METHOD: The phagecytosis of murine macrophages was analyzed by neutral red phagecytosis assay. The activities of IL-1 and TNF-alpha were measured by biological methods. The mRNA of TNF-alpha and INF-gamma expressed by macrophages was detected by RT-PCR. RESULT: The phagecytosis of murine macrophages was significantly enhanced by FLM at a concentration from 1 microg x mL(-1) to 100 microg x mL(-1) and the secretions of IL-1, and TNF-alpha from macrophages were markedly induced by FLM. Meanwhile, FLM also increased the expression of TNF-alpha mRNA and INF-gamma mRHA from macrophages in vitro. CONCLUSION: FLM could promote phagecytosis and cytokines production of murine macrophages.

Abstract Ethnopharmacological relevance Zuotai (gTso thal) has a long history in the treatment of cardiovascular disease, liver and bile diseases, spleen and stomach diseases as a precious adjuvant in Tibetan medicine. However, Zuotai is a mercury preparation that contains 54.5% HgS. Its application has always been controversial. Aim of the study To evaluate the toxicological effects of Zuotai in hepatocytes and in zebrafish. Materials and methods MTT was used to determine the survival rate of hepatocytes; Hoechst and TUNEL staining were used to detect the apoptosis cells; Western blot and RT-qPCR assay were used to determine the expression levels of the protein and mRNA; Liver morphology observation and H&E staining were used to evaluate the hepatotoxicity of Zuotai in Zebfrafish. Results The survival rate of L-02 cells, HepG2 cells and RBL-2A cells reduced by Zuotai (10−4–0.1 mg/mL) in a dose and time-dependent manner. Zuotai (0.1 mg/mL) induced HepG2 cells shrinkage, condensation and fragmentation and increased the number of apoptosis cells. The protein expression levels of cleaved Caspase-3 and Bax were increased and the expression levels of Bcl-2 were reduced after HepG2 cells exposed to Zuotai (10−4–0.1 mg/mL) for 24 h. In addition, Zuotai (0.2 mg/mL) induced the darker liver color of the larval zebrafish and changed the liver morphologic of adult zebrafish. Zuotai (0.2 mg/mL) also increased the mRNA levels of CYP1A1, CYP1B1 and MT-1 in the liver of adult zebrafish. However, no significantly hepatotoxicity was observed after hepatocytes and zebrafish exposed to HgS at the same dose. Conclusions Results showed that Zuotai induced hepatotoxicity effectively under a certain dose but its hepatotoxicity likely occurs via other mechanisms that did not depend on HgS. BACKGROUND: As drug use has limitations in the treatment of irritable bowel syndrome (IBS), increasing attention is being paid to nondrug therapies and complementary treatments, especially exercise. It is known that bowel movements are more frequent and colon transit is more rapid in physically active individuals than in sedentary individuals. However, the effects of exercise on IBS are unclear. PURPOSE: We conducted a systematic review to assess the effects of exercise on IBS. METHODS: We searched PubMed, Web of Science, EMBASE, Cochrane Library, and two Chinese databases (Wanfang Database and Chinese Biomedical Literature [CBM]) for eligible studies. We extracted and pooled relevant information regarding the effects of exercise in patients with IBS. The primary outcomes were gastrointestinal symptoms, quality of life, depression, and anxiety. KEY RESULTS: A total of 683 patients with IBS from 14 randomized controlled trials met our inclusion criteria. The exercise interventions in this review were yoga, walking/aerobic physical activity, Tai Ji, mountaineering, and Baduanjin qigong activity. The results of this review suggested exercise had significant benefits for patients with IBS, but studies were limited by the strong risk of bias. CONCLUSIONS AND INFERENCES: Our findings suggest that exercise is potentially a feasible and effective treatment for IBS patients. However, because of methodologic issues, no firm conclusions could be drawn about the true effects of this intervention. Researchers should design a rigorous study to assess the effects of exercise on IBS.
