The aim of this study was to investigate the antidiabetic effects of a Tibetan medicine, Tang-Kang-Fu-San (TKFS), on experimental type 2 diabetes mellitus (T2DM) rats and to explore its underlying mechanisms. Firstly two major chemical compositions of TKFS, gallic acid and curcumin, were characterized by HPLC fingerprint analysis. Next T2DM in rats was induced by high-fat diet and a low-dose streptozotocin (STZ 35 mg/kg). Then oral gavage administration of three different doses of TKFS (0.3 g/kg, 0.6 g/kg, and 1.2 g/kg) was given to T2DM rats. Experimental results showed that TKFS dramatically reduced the levels of fasting blood glucose, fasting blood insulin, triglyceride, total cholesterol, LDL cholesterol, and HDL cholesterol, even though it did not alter the animal body weight. The downregulation of phosphorylation-AKT (p-AKT) and glucose transporter-4 (GLUT4) in skeletal muscle of T2DM rats was restored and abnormal pathological changes in pancreas tissues were also improved. Our work showed that TKFS could alleviate diabetic syndromes, maintain the glucose homeostasis, and protect against insulin resistance in T2DM rats, and the improvement of AKT phosphorylation and GLUT4 translocation in skeletal muscle would be one of its possible underlying mechanisms.
The current study evaluated antioxidant activity of <i>Dracocephalum rupestre</i> Hance, characterized by the polyphenolic compounds in the ethyl acetate fraction (EAF) from <i>D. rupestre</i> and investigated the protective mechanisms of EAF in carbon tetrachloride (CCl₄)-induced hepatic injury. EAF showed the largest antioxidant capacity as demonstrated by DPPH, ABTS, OH, and FRAP assays (<i>p</i> ≤ .05). Using LC-MS, two polyphenolics, for example, rosmarinic acid, and eriodictyol were identified in EAF. Total phenol content of <i>D. rupestre</i> was correlated significantly with FRAP value (<i>r</i> = .999, <i>p</i> ≤ .001). The results also showed that EAF decreased serum ALT and AST activities compared with model group, as well as the histological findings. In addition, EAF remarkably decreased MDA and LDH levels in liver. This is the first time to reveal the hepatoprotective effect of EAF from <i>D. rupestre</i>, which may be developed as a new drug for treatment of liver injury in future.<br>Practical applications: <i>Dracocephalum rupestre</i> Hance is known as Chinese herbal medicine with pharmacological functions of antioxidant, anti-inflammatory and has been mainly used in the treatment of damp-heat, headache, fever, jaundice, hepatitis, liver toxicity, and other diseases. In order to detect the potential application of this plant, the antioxidant and hepatoprotective activities of <i>D. rupestre</i> were investigated. It was found that the ethyl acetate fraction (EAF) from <i>D. rupestre</i> displayed strong antioxidant activity. We also demonstrate that EAF could lessen CCl₄-induced acute liver injury in mice. Also, rosmarinic acid may play an important role in the antioxidant activity and hepatoprotective effect for EAF.
In this paper, an efficient method was successfully established by the combination of macroporous resin (MR) and high-speed counter-current chromatography (HSCCC) for rapid enrichment and separation of aloe-emodin 8-O-β-D-glucoside, emodin 1-O-β-D-glucoside, emodin 8-O-β-D-glucoside and piceatannol 4'-O-β-D-(6″-O-gallate)-glucoside. Six kinds of macroporous resins were investigated in the first step and X-5 macroporous resin was selected for the enrichment of the target compounds. The recoveries of the target compounds reached 89.0, 85.9, 82.3 and 84.9% respectively after 40% ethanol elution. In the second step, the target compounds were separated by HSCCC with a two-phase solvent system composed of chloroform/ethyl acetate/methanol/water (8:1:6:5, v/v). The established method will be helpful for further characterization and utilization of Rheum tanguticum. The results demonstrate that MR coupled with HSCCC is a powerful technique for separation of bioactive compounds from natural products.
Background: Veronica ciliata Fisch., widely distributed in western China, has been traditionally used in Tibetan Medicine as a treatment for hepatitis, cholecystitis, rheumatism, and urticaria. However, V. ciliata Fisch. has not been subjected to detailed chemical constitution analysis and the bioactive studies were restricted to its crude extracts. It is necessary to investigate the active chemical components of these extracts and identify their biological effects. Results: Four iridoid glycosides, (veronicoside, cataposide, amphicoside, and verminoside) were isolated from the ethyl acetate fraction. Among these compounds, veronicoside and verminoside were isolated for the first time from this plant. These compounds exhibited strong antioxidant activity and inhibitory activity on HepG2 cell proliferation. The antioxidant activity of verminoside was equal to Vc. Cataposide, amphicoside and verminoside had stronger anti-hepatocarcinoma activity than 5-fluorouracil. Conclusions: Four iridoid glycosides,(veronicoside, cataposide, amphicoside and verminoside) were isolated from the extract of V. ciliata Fisch. using bioassay-guided screening.Among these compounds, veronicoside and verminoside were isolated for the first time from this plant. The above results indicated that these compounds were the active chemical components responsible for the antioxidant and anti-hepatocarcinoma properties of V. ciliata Fisch. The underlying mechanism of their bioactivity is worthy of further investigation. [Figure not available: see fulltext. To induce callus and establish rapid propagation systems for Veronica ciliate Fisch., terminal buds from its aseptic seedlings were cultured in MS mediums with different concentration combinations of NAA and 6-BA. Results showed that the optimum medium for callus inducement was MS + 6-BA 0. 5 mg/L + NAA 1. 0 mg/L, with the callus induction rate of 95%. The best medium for multiplication was MS + 6-BA 0. 5 mg/L + NAA 0. 1 mg/L,with the multiplication coefficient reaching 5. 4. The optimum root medium for multiple shoot clumps was 1/2 MS + IBA 0. 05 mg/L,with the rooting rate reaching 95% and the root being healthiest. Under suitable hardening and transplanting conditions, the highest survival rate for the regenerated plantlets reached 40%.
Five days of integrative body-mind training (IBMT) improves attention and self-regulation in comparison with the same amount of relaxation training. This paper explores the underlying mechanisms of this finding. We measured the physiological and brain changes at rest before, during, and after 5 days of IBMT and relaxation training. During and after training, the IBMT group showed significantly better physiological reactions in heart rate, respiratory amplitude and rate, and skin conductance response (SCR) than the relaxation control. Differences in heart rate variability (HRV) and EEG power suggested greater involvement of the autonomic nervous system (ANS) in the IBMT group during and after training. Imaging data demonstrated stronger subgenual and adjacent ventral anterior cingulate cortex (ACC) activity in the IBMT group. Frontal midline ACC theta was correlated with highfrequency HRV, suggesting control by the ACC over parasympathetic activity. These results indicate that after 5 days of training, the IBMT group shows better regulation of the ANS by a ventral midfrontal brain system than does the relaxation group. This changed state probably reflects training in the coordination of body and mind given in the IBMT but not in the control group. These results could be useful in the design of further specific interventions.

BACKGROUND:Psychological distress and metabolic dysregulation are associated with markers of accelerated cellular aging, including reduced telomerase activity and shortened telomere length. We examined whether participation in a mindfulness-based intervention, and, secondarily, improvements in psychological distress, eating behavior, and metabolic factors are associated with increases in telomerase activity in peripheral blood mononuclear cells (PBMCs).
METHODS:
We enrolled 47 overweight/obese women in a randomized waitlist-controlled pilot trial (n=47) of a mindfulness-based intervention for stress eating and examined changes in telomerase activity from pre- to post-intervention. In secondary analyses, changes in telomerase activity across the sample were examined in relation to pre- to post-intervention changes in psychological distress, eating behavior, and metabolic factors (weight, serum cortisol, fasting glucose and insulin, and insulin resistance).
RESULTS:
Both groups increased in mean telomerase activity over 4 months in intent-to-treat and treatment efficacy analyses (p<0.001). Nonsignificant trends showed that greater attendance was associated with increases in telomerase, and telomerase increases were 18% higher among 'as treated' participants compared to controls. Across groups, changes in chronic stress, anxiety, dietary restraint, dietary fat intake, cortisol, and glucose were negatively correlated with changes in telomerase activity. In exploratory analyses, decreases in dietary fat intake partially mediated the association between dietary restraint and telomerase activity with marginal significance.
CONCLUSIONS:
While there was no clear effect of the intervention on telomerase activity, there was a striking pattern of correlations between improvements in psychological distress, eating behavior, and metabolic health and increases in telomerase activity. These findings suggest that telomerase activity may be in part regulated by levels of both psychological and metabolic stress.
In this chapter, Lin and Fyles retrace the thinking of Aldo Leopold on conceiving humans as plain members and citizens of the biotic community, perceiving the intrinsic character of the land as “land health,” and considering how these two ideas led to his famous “land ethic.” Leopold’s land ethic urges people to expand their relationship with land beyond economics to include “integrity, stability, and beauty.” The authors then apply his thinking to the context of ecosystem and ecosystem health by considering humans as part of ecosystems, and the resulting implications for ecosystem health. With this thinking, the authors return to Leopold by studying one of his essays in A Sand County Almanac (1949). In “A Mighty Fortress,” Leopold mentioned how his woodlot, having been visited by various tree diseases, became a rich habitat for wildlife. This essay underscores the multiplicity of perspectives in an ecosystem and its complex nature, which in turn challenges humans to learn the richness and meaning of the concept of health. Put another way, and echoing the view of the previous chapter, health cannot be adequately portrayed by using only medical science. The authors conclude with a call for supplementing the scientific, rational mode of perceiving reality and human action with thinking from the arts and the humanities.
In order to reveal the chemical substance basis of pharmacodynamic effects of Zuotai, energy dispersive spectrometry of X-ray (EDX), X-ray fluorescence spectroscopy (XRF), synchrotron radiation X-ray absorption fine structure (SR-XAFS), X-ray diffraction (XRD), scanning electron microscope (SEM) and atomic force microscope (AFM) were used to analyze the elements, the chemical valence and local structure of mercury, and the chemical phase composition and micro-morphology of Zuotai. EDX and XRF analysis shows that the main elements in Zuotai are Hg and S, with some other minor elements, such as 0, Fe, Al, Cu, K, Ag, Ca, Mg etc. SR-XAFS analysis shows that: the oxidation state of mercury in Zuotai is divalence, its neighbor atoms are S, and its coordination number is four. XRD assay found that β-HgS (cubic, F-43m 216) and S8 (orthorhombic, Fddd 70) are the main phase compositions in Zuotai. Besides, it also has a small amount of C (hexagonal, P63/mmc 194), Fel.05 S0.95 (hexagonal, P63/mmc 194), Cu6S6 (hexagonal, P63/mmc 194), Cu1.8 S (cubic, F-43m 216) and so on. And it was found that the crystallinity of Zuotai is about 59%, and the amorphous morphology substance in it is about 41%. SEM and AFM detection suggests that Zuotai is a kind of ancient micro-nano drug, and its particle size is mainly in the range of 100-600 nm, even less than 100 nm, which commonly further aggregate into several to 30 µm loose amorphous particles. In summary, the present study elucidated physicochemical characterization(elements composition, coordination information of mercury, phase composition and micro-morphology) of Zuotai, and it will play a positive role in promoting the interpretation of this mysterious drug.;
OBJECTIVE: To study the chemical constituents of Arenaria kansuensis.METHOD: Column chromatographic techniques were applied to isolated constituents. A combination of physico-chemical properties and spectroscopic analysis were used to identify structures of constituents.
RESULT: Five compounds were obtained and elucidated as tricin (1), tricin-7-O-beta-D-glucopyranoside (2), tricin 4'-O-beta-guaiacylglyceryl ether (3), isoscoparin (4) and isovitexin (5).
CONCLUSION: Compound 2, 3 and 5 were isolated from A. kansuensis for the first time, compound 3 was obtained from the family Caryophyllaceae for the first time.
The seeds of Herpetospermum caudigerum are used in the traditional Tibetan medicine for the treatment of liver diseases.
OBJECTIVE: To compare the ettect and side-ettect of fluoxetine and combination of fluoxetine and Chinese or Tibetan medicine in treating senile depression in plateau district. Methods Ninety patients with diagnosis of senile depression conformed to CCMD-3 standard, in plateau district of 2260 - 3200 m altitude were randomly divided into three groups and treated with fluoxetine (group A), fluoxetine plus Sanpu Xinnao Xin granule (group B) and fluoxetine plus Xiaoyao pill (group C), respectively, 30 cases in each group. Therapeutic effects were evaluated with Hamilton' s depressive scale (HAMD) and treatment emergent symptom scale (TESS) after 6 weeks treatment.RESULTS: There was no significant difference in the therapeutic effects between the three groups. The adverse reaction in Group B and C was less than that in Group A (P<0.01). Conclusion Sanpu Xinnao Xin granule and Xiaoyao pill can raise the tolerance of patients with senile depression in plateau area against the adverse reaction of fluoxetine.
In an effort to discover potent VEGFR-2 inhibitors, a series of 2,4 or 4,6-disubstituted <b>O</b>-linked indoles derivatives were designed and synthesized. The structural activity relationships led to identification of a potential VEGFR-2 inhibitor compound <b>18</b>.<br>Inhibition of VEGFR-2 signaling pathway has already become one of the most promising approaches for the treatment of cancer. In this study, we describe the design, synthesis, and biological evaluation of a series of <b>O</b>-linked indoles as potent inhibitors of VEGFR-2. Among these compounds, <b>18</b> showed significant anti-angiogenesis activities <b>via</b> VEGFR-2 in enzymatic proliferation assays, with IC50 value of 3.8 nmol/L. Kinase selectivity profiling revealed that <b>18</b> was a multitargeted inhibitor, and it also exhibited good potency against VEGFR-1, PDGFR-<b>α</b> and <b>β</b>.
A new RP-HPLC method was developed for the simultaneous determination of 5 active constituents, including loganic acid, swertiamarin, gentiopicroside, sweroside, isoorientin in aerial part of Gentiana straminea. Analysis was achieved on a Hypersil ODS analytical column (4.6 mm x 250 mm, 5 microm) eluted with methanol and water containing 0.02% phosphoric acid in gradient elution. The flow rate was 1 mL x min(-1), the column temperature was set at 35 degrees C and detection wavelength was set at 254 nm. The results showed that 5 active components were separated well and showed good linearity. The corelation coefficints and concentration ranges of the calibration curves were as follow: r = 0.999 9, 0.364-2.18 microg for loganic acid. r = 0.999 9, 0.275-1.65 (microg for swertiamarin. r = 0.999 9, 0.614-3.68 microg for gentiopicroside. r = 0.999 9, 0.065 6-0.394 microg for sweroside. r = 0.999 9, 0.089 9-0.539 microg for isoorientin. The contents of loganic acid and gentiopicroside far exceed that of the other 3 active components, and made up 60% in the total contents. The developed method was proved to be rapid, sensitive, accurate, credible and repeatable. It can be applied to quality control of aerial part of Tibetan medicine Gentiana straminea.
Honghua Ruyi Wan (HHRYW) is a traditional Tibetan drug. We designed this study to investigate the effectiveness of HHRYW for relief of pelvic inflammatory disease (PID) pain in women. We performed a multicenter, randomized, double-blind, placebo-controlled trial, and observed the effectiveness and toxicity of HHRYW in combination with moxifloxacin in treating PID. Of the 139 women enrolled in this study, 124 were included in the final analysis. They were divided into the HHRYW group (n=65) and the placebo group (n=59). The baseline age, height, weight, and marital status were well matched between the groups (all: P>0.05). The rate of reduction in tenderness score was significantly greater in the HHRYW group than in the placebo group after 30, 60, and 90 days of treatment. The HHRYW group had a significantly lower subjective visual analogue scale score at 30, 60, and 90 days after treatment. The incidence of adverse reactions was 7.04% (5/71) in the HHRYW group and 10.61% (7/59) in the placebo group. No severe adverse reactions were noted in either group. The combination of HHRYW and antibiotics effectively relieve abdominal pain caused by PID with obvious long-term efficacy and acceptable adverse reactions. [ABSTRACT FROM AUTHOR]
This study aimed to find a simple and effective mindfulness training method for intervening depression. 95 university students with moderate or severe depression participated in the study and they were divided into three mindfulness training groups: 8-week complete group, 8-week informal group, and 4-week group. All subjects completed Beck Depression Inventory (BDI-Ⅱ) and Five Fact Mindfulness Questionnaire (FFMQ) before the intervention, after which, 55 subjects completed the assessments once more. Results showed the scores of specific dimensions representing mindfulness levels were different. For the 4-week group, scores of description, action aware and nonjudge significantly were improved, while the other two groups were improved significantly on all the five dimensions, especially the informal training group. In the discussion, it focused more on the effects of training time and training contents, and briefly discussed the relation of subjects dropout, subjects attendance, homework completion status and intervention effects. In conclusion, all the three intervention modes have magnificent intervention effects. Integrated many sided, 8-week informal training is of the best effects, which may be considered as a better mindfulness training mode henceforth, however, the comparison between the formal training and informal training still needs further study.

The efficacy of a single-session Hatha yoga class on stress reduction is not currently known.PURPOSE: This study investigated the comparative effectiveness of a single 90-minute Hatha yoga class and an 8-week, 90-minute-class-per-week course.
METHODS: We used a quasiexperimental design and recruited 63 female community residents in New Taipei City aged 40-60 years. Participants were randomly divided into an experimental group (n = 30) and a control group (n = 33). The experimental group received the 8-week Hatha yoga course. The control group received no intervention. The Perceived Stress Scale (PSS) and heart rate variability (HRV) assessed stress reduction effectiveness. Chi-square, independent t test, paired t test, and generalized estimating equations were used for data analysis.
RESULTS: After a single 90-minute class of Hatha yoga, experimental group PSS scores were significantly less than those of the control group (p = .001). Although experimental group HRV (low-frequency norm and high-frequency norm) had improved, these changes were not statistically significant (p = .059). PSS scores for the single 90-minute class and 8-week course did not significantly differ (p = .157) and HRV of statistics is significant (p = .005). Generalized estimating equations analyzed changes in the effectiveness over time of stress reduction (HRV and PSS) after the Hatha yoga intervention. Results showed the postintervention HRV and PSS of the experimental group decreased significantly (p < .001) more than the control group.
CONCLUSIONS/IMPLICATIONS FOR PRACTICE: Our findings support the position that regular, long-term practice of Hatha yoga provides clear and significant health benefits. Objective: Prehypertension is a new category designated by the Seventh Report of the Joint National Committee on Prevention, Detection, Evaluation, and Treatment of High Blood Pressure ( JNC7) in 2003. Managing prehypertension with nonpharmacological intervention is possibly beneficial to the prevention of hypertension. In this study, we observed the effect of slow abdominal breathing combined with electromyographic (EMG) biofeedback training on blood pressure (BP) in prehypertensives and assessed the changes of heart rate variability (HRV) in order to find an optional intervention to prevent hypertension and acquire some experimental data to clarify the underlying neural mechanism.Methods: Twenty-two (22) postmenopausal women with prehypertension were randomly assigned to either the experiment group or the control group. The experiment group performed 10 sessions of slow abdominal breathing (six cycles/min) combined with frontal electromyographic (EMG) biofeedback training and daily home practice, while the control group only performed slow abdominal breathing and daily home practice. BP and HRV (including R–R interval and standard deviation of the normal–normal intervals [SDNN]) were measured.
Results: Participants with prehypertension could lower their systolic blood pressure (SBP) 8.4 mm Hg ( p < 0.001) and diastolic blood pressure (DBP) 3.9 mm Hg ( p < 0.05) using slow abdominal breathing combined with EMG biofeedback. The slow abdominal breathing also significantly decreased the SBP 4.3 mm Hg ( p < 0.05), while it had no effect on the DBP ( p > 0.05). Repeated-measures analyses showed that the biofeedback group þ abdominal respiratory group (ABþBF) training was more effective in lowering the BP than the slow breathing ( p < 0.05). Compared with the control group, the R–R interval increased significantly during the training in the ABþBF group ( p < 0.05). The SDNN increased remarkably in both groups during the training ( p < 0.05).
Conclusions: Slow abdominal breathing combined with EMG biofeedback is an effective intervention to manage prehypertension. The possible mechanism is that slow abdominal breathing combined with EMG biofeedback could reduce sympathetic activity and meanwhile could enhance vagal activity.
The present study investigated changes in autonomic nervous system activity and emotions after a short (2 h) forest bathing program in the Xitou Nature Education Area (XNEA), Taiwan. One hundred and twenty-eight (60.0 ± 7.44 years) middle-aged and elderly participants were recruited. Physiological responses, pulse rate, systolic and diastolic blood pressure, heart rate variability (HRV), and psychological indices were measured before and after the program. We observed that pulse rate, systolic and diastolic blood pressure were significantly lower after the program, which indicated physiological benefits from stress recovery. The Profile of Mood States negative mood subscale scores of “tension-anxiety”, “anger-hostility”, “fatigue-inertia”, “depression-dejection”, and “confusion-bewilderment” were significantly lower, whereas the positive mood subscale score of “vigor-activity” was higher. Furthermore, participants exhibited significantly lower anxiety levels according to the State-Trait Anxiety Inventory. However, changes in sympathetic and parasympathetic nerve activity were nonsignificant. Our study determined that the short forest bathing program is a promising therapeutic method for enhancing heart rate and blood pressure functions as well as an effective psychological relaxation strategy for middle-aged and elderly individuals.
PURPOSE: Breast cancer is the most common female cancer. The aim of this study was to assess the impact of yoga on lymphedema in breast cancer survivors.DESIGN: Repeated measures before and after the intervention. We enrolled 15 women with breast cancer who had not previously worn elastic clothing to treat lymphedema.
METHODS: The program was led by a certified trainer and consisted of 60-minute sessions, three times a week for 12 weeks. The volumes of the affected and normal limbs were measured. A self-assessed edema score was also recorded.
FINDINGS: Fifteen patients completed the program, none of whom suffered from complications related to exercise. There was no significant edema after exercise. No significant differences were noted in subgroup analysis by age or the affected arm.
CONCLUSIONS: Yoga does not induce lymphedema.
CLINICAL RELEVANCE: Lymphedema is usually treated with uncomfortable elastic clothing, and high-resistance exercise may induce edema. Yoga may be suitable for these patients.
